What Is AOD-9604?
AOD-9604 (Anti-Obesity Drug 9604) is a synthetic modified fragment corresponding to the C-terminal region of human growth hormone — residues 176-191 of the 191-amino-acid hGH molecule, with an added disulfide bridge. It was originally developed in the 1990s at Monash University as an anti-obesity candidate and later advanced through clinical development by Metabolic Pharmaceuticals.
The proposed mechanism links AOD-9604 to beta-3 adrenergic receptor activation, which in preclinical models is associated with increased lipolysis (breakdown of stored fat) and decreased lipogenesis (formation of new fat). Because it lacks the receptor-binding regions of full-length GH, it is not expected to produce the growth-promoting, IGF-1-raising, or blood-sugar effects associated with exogenous GH administration.
AOD-9604 completed six placebo-controlled clinical trials with over 900 combined participants. Development for the obesity indication was discontinued in 2007 after the largest Phase IIb trial failed to show statistically significant weight loss versus placebo at the primary endpoint.
